Ryanair will open a new base at Prague Airport on October 30, with two aircraft.
Alongside its existing routes to Brussels, Dublin and London, it will also fly from Prague to Milan and Rome.
This will bring the total number of weekly flights to over 40.
Dublin flights operate daily and London Stansted twice a day.
"This growth is an integral part of our continued expansion plans for both the Czech Republic and Eastern Europe," said Kenny Jacobs.
